چکیده

The performance of seed tubers harvested from potato plants grown from sprout cutting, stem cutting and conventional seed tubers against PVY and PLRV were investigated. Sprout cutting, stem cutting and conventional seed tubers of variety Diamant were planted and second and third generation seed tubers were harvested. The least incidence of PVY and PLRV was recorded when seed tubers from sprout cutting were used. Whereas, the highest incidence of the viruses was observed where conventional seed tubers wete planted. Incidence of PVY, PLRV and their mixed infection was lower in earlier generations as compared to later generations. Performance of seed tubers produced from sprout cutting was better in respect of plant growth, tuber number and tuber yield per hill as compared to seed tubers obtained from stem cutting or conventional method.

Infection Pattern of PVY and PLRV in Potato Tubers Obtained from Potato Plants Infected with Both the Viruses

Experiments were conducted to study the plant growth and tuber yield of the seed potato collected from PVY and PLRV infected plants. The seed potatoes were graded as under size (<28 mm), A grade ( 28-40 mm), B grade (41-55 mm) and over size (>55 mm) which were planted following RCBD in the field. Third generation seed potatoes of Diamant variety were used. The incidence of viruses was confirmed...

Digoxigenin-labelled molecular probe for the simultaneous detection of three potato pathogens: potato spindle tuber viroid (PSTVd), potato virus Y (PVY), and potato leafroll virus (PLRV).

A molecular probe, p3POT, was constructed of PSTVd, PVY, PLRV cDNA fragments introduced into pUC18 vector. Sequencing of the inserts revealed that cloned fragments covered conservative parts of pathogenic genomes. Dot-blot hybridization of digoxigenin-labelled construct to crude extracts from plants infected with different potato viruses proved high sensitivity and specificity of the p3POT prob...

Effect of Seed Size and Cutting Methods on the Yield and Profitability of Potato

An experiment was conducted at the research field of Tuber Crop Research Centre (TCRC) of Bangladesh Agricultural Research Institute, Gazipur during the winter of 2006-2007 to determine the effects of cutting methods and seed size on the yield and profitability in potato (Solanum tuberosum L.). It was observed that the foliage coverage, plant height, stems/hill, number and weight of tubers/plan...

Induction mechanism of 3-hydroxy-3-methylglutaryl-CoA reductase in potato tuber and sweet potato root tissues.

3-Hydroxy-3-methylglutaryl coenzyme A reductase (HMGR, EC1.1.1.34), the key enzyme in isoprenoid biosynthesis, was purified from microsomes of potato tuber tissue, and a polyclonal antibody and two monoclonal antibodies against the purified enzyme were prepared. HMGR protein content was measured by immunotitration and radioimmunoassay using these antibodies. HMGR activity was very low in the fr...
